<p>Just like in qPCR experiments, sample preparation includes the transfer of master mix, probes and primers to a 96- or 24-well nanoplate, followed by the addition of samples. The system integrates partitioning, thermocycling and imaging into a single fully automated instrument that takes users from the sample to result in under 2 hours. One can perform analysis on the Software Suite, providing the concentration in copies per microliter of your target sequence as well as for quality control such as positive samples or NTC. This analysis can also be extended to remote computers within the same local area network (LAN).</p>

<p>The QIAcuity Software Suite provided with the instrument and installed on a separate computer controls one or multiple QIAcuity instruments, either connected directly to one instrument or using an existing local area network (LAN). Using the QIAcuity Software Suite, digital PCR experiments, samples and reaction mixes can be defined, assigned to Nanoplates, and transferred to the QIAcuity instrument. After the run, data can be analyzed, reports can be created and data can be exported for external analysis. The software provides multiple template functionalities to make repeating plate layouts or plate run parameters easily accessible, further transforming your digital PCR experience.</p>
<p>When integrated into a local area network, the computer is hosting the QIAcuity Software Suite function as a server that is accessible via LAN to other computers serving as clients. Simply place samples and buffers into the instrument and start the run. The tips of the gel cartridges move into the sample wells. An electric field creates a positive charge. This makes the negatively charged nucleic acid fragments migrate through the gel in the capillaries, with the smaller fragments travelling faster than the larger ones. Within the capillary, a dye intercalates into the nucleic acid. As the fragments pass a detector, it measures the signals emitted by the dye. The QIAxcel ScreenGel software converts these signals to digital data; you can see them in real time as electropherograms or gel images (see Cartridge-based gel electrophoresis). High performance PCR setup using QIAgility A QuantiNova SYBR Green PCR assay setup was performed manually and automated in parallel using the QIAgility along with its Setup Manager Software. The amplification run was done on the Rotor-Gene Q and resulting data analyzed using the Q-Rex Software. Consistent results were obtained from both manual and automated reaction setup using the same template dilutions (see figure “ Comparison of manual and automated assay setup using series dilutions”). Exceptional linearity of QIAgility QIAgility exhibits excellent linearity and assures accurate and reliable use of sample for an assay. To assess the linearity of the system, pure gDNA was serially diluted using the QIAgility and measured for their concentrations on the QIAxpert. The quantifications from the QIAxpert were plotted against the known concentration values, and the resulting plot demonstrates the exceptional linearity (with a regression coefficient of &gt;0.999) of QIAgility (see figure “ High system linearity of QIAgility”). Principle Unique rotary design for outstanding performance The Rotor-Gene Q MDx uses a sophisticated heating and cooling design to achieve optimal reaction conditions. The unique rotary format ensures optimal thermal and optical uniformity between samples, which is critical for precise and reliable analysis (see figure " Cross-section of the Rotor-Gene Q MDx").
